# Advancements in RNA therapeutic technologies

Developments in RNA technology are expanding from messenger RNA (mRNA) applications toward new therapeutic frontiers, including transfer RNA (tRNA). 

In oncology, personalized mRNA cancer vaccines are being tested in combination with immunotherapy. Clinical trials involving melanoma patients suggest that sequencing specific tumor mutations to train the immune system may reduce the risk of recurrence and metastasis. This approach is being considered for other high-mutation cancers, such as lung and colon cancer.

Simultaneously, research is advancing the use of tRNA to address hereditary diseases caused by nonsense mutations. By utilizing ‘suppressor tRNAs’ to bypass premature stop signals during protein synthesis, researchers aim to correct the functional impact of genetic errors. This technology holds potential for treating conditions such as cystic fibrosis, Duchenne muscular dystrophy, and Dravet syndrome.

Recent progress includes a next-generation tRNA approach developed at the University of Toronto, where researchers demonstrated the ability to restore protein production in preclinical models of cystic fibrosis by engineering tRNA to read through premature stop signals. 

Additionally, efforts to streamline mRNA drug development are underway at SINTEF. To address the high failure rate of drugs in human clinical trials caused by discrepancies between animal models and humans, a new method utilizing human blood is being developed. This approach aims to identify effective drug candidates earlier in the process for mRNA treatments targeting cardiovascular diseases, autoimmune disorders, and cancer.
