# MIT research into lung cancer enzymatic targets

Researchers at the Massachusetts Institute of Technology (MIT) have identified enzymatic mechanisms that could facilitate ‘cancer interception’, a method of intervening before tumors become established.

Initial findings published in Science Advances highlighted the inflammatory enzyme caspase-1 as a potential target. In studies involving mice, blocking this enzyme significantly reduced the growth and development of lung tumors. Lead author Sangeeta Bhatia suggested that oral small-molecule caspase-1 inhibitors could provide a convenient long-term preventive treatment for high-risk individuals.

Subsequent research expanded on these findings by identifying additional drivers of lung cancer. A study in Nature Communications identified the enzyme UCHL1 as a critical factor in lung adenocarcinoma. UCHL1 functions as a deubiquitinating enzyme that stabilizes proteins necessary for cancer cell survival, allowing malignant cells to maintain high metabolic rates and bypass natural cell death. 

Further laboratory experiments demonstrated that combining a caspase-1 inhibitor with an antibody against IL-1 beta allowed nearly 20 percent of mice to avoid tumor development entirely. These developments suggest that targeting specific inflammatory and metabolic pathways may reduce tumor progression while minimizing damage to healthy tissue.
