Researchers at Nagoya University have developed a method to create pituitary-hypothalamic organoids using human embryonic stem cells. These three-dimensional structures are capable of producing adrenocorticotropic hormone (ACTH).

Initial studies demonstrated that transplanting these organoids into mice with removed pituitary glands increased ACTH concentrations and improved survival rates over six months without uncontrolled cell growth. The research subsequently progressed to a 15-year-old Javan macaque that had undergone surgical removal of its pituitary gland.

In the primate study, the transplant enabled the recovery of essential hormonal signals, specifically increasing cortisol levels. This development is significant for regenerative medicine, as it suggests a potential future alternative to lifelong hormone replacement therapy, which currently struggles to mimic the body’s natural regulatory rhythms.
