Research into neurodegeneration has highlighted the role of the peripheral immune system in driving brain damage. Initial studies from the Washington University School of Medicine indicate that CD8+ T cells, which contribute to inflammation and brain damage in Alzheimer’s disease, may be “instructed” in peripheral lymphatic tissues, such as deep cervical lymph nodes, rather than within the central nervous system.

Subsequent findings using murine models of tauopathy support this connection, suggesting that the mechanisms driving Alzheimer’s and other tauopathies may originate outside the brain. Researchers observed that mice lacking specific dendritic cells or the ability to present certain antigens to CD8+ lymphocytes showed significant protection from neurodegenerative damage. In these protected models, fewer CD8+ lymphocytes reached the brain and there was lower activation of glial cells, suggesting that targeting the peripheral immune system could offer new therapeutic avenues.
