Advancements in CAR-T cell therapy research and effectiveness
Recent research highlights significant developments in the engineering and clinical observation of Chimeric Antigen Receptor (CAR)-T cell therapies for cancer treatment.
One study introduced a novel cloning strategy using barcoded protein domain combinations to expedite the development of CAR constructs. By assembling individual domains with unique DNA barcodes, researchers generated 360 constructs targeting CD19 and GD2 antigens. The screening identified that hinge domains, specifically CD8a and IgG4, significantly influence surface expression and antitumor activity. Notably, the study discovered two CD19-CAR architectures using the IgG4 hinge that demonstrated superior in vivo antitumor activity compared to the FDA-approved therapy Kymriah.
In a separate observational study involving 715 patients, researchers investigated the diurnal rhythm of CAR-T cell effectiveness. This study aims to understand how the timing of treatment or biological rhythms may impact the efficacy of these immunotherapy cells.
