Alzheimer's studies reveal neuronal survival mechanism and new cell‑death pathway
Researchers at the Netherlands Institute for Neuroscience reported that about 30 % of older adults who develop Alzheimer’s pathology remain cognitively intact. Their brains contain fewer immature neurons, but the surviving neuroblasts display a distinct transcriptional profile that supports their survival and maturation despite the disease environment. The authors suggest that boosting the behavior of these cells could become a therapeutic strategy for the majority who develop dementia.
A separate study by King’s College London identified a novel form of neuron loss called karyoptosis, in which the nucleus disintegrates. The process is driven by the enzyme p38 MAPK tagging the structural protein LaminB1 for degradation. Blocking p38 MAPK delayed nuclear collapse in laboratory neurons, even though toxic protein levels remained unchanged. Analysis of post‑mortem brain tissue showed karyoptosis in 35 % of cortical cells from Alzheimer’s and frontotemporal dementia patients versus 15 % in healthy controls, highlighting a potential target for future treatments.
