Biological mechanisms link stress and trauma to psychiatric vulnerability

Researchers have identified biological mechanisms that explain how stress and trauma impact brain function and increase vulnerability to psychiatric disorders.

A study conducted by the University of Sydney and the Max Planck Institute of Psychiatry found that in individuals with schizophrenia, the FKBP5 gene—which regulates the body’s response to stress hormones like cortisol—is more easily activated. Using donated brain tissue, researchers observed that the chemical tags that typically regulate this gene are stripped away, effectively leaving the stress response “stuck on high.” This heightened reactivity can increase vulnerability to psychiatric conditions, particularly as the effects become more pronounced with age.

In a separate study involving Washington University School of Medicine and Princeton University, scientists discovered how childhood trauma can leave a lasting biological “scar” on brain cells. The research suggests that early-life adversity alters how brain cells package their DNA, making stress-related genes easier to activate. This process, particularly within the ventral tegmental area responsible for dopamine production, can leave the brain more reactive and less able to tolerate future stress, contributing to long-term risks for anxiety, depression, and other mood disorders.
