Alzheimer's biomarkers detectable years before symptoms, studies show

Large‑scale studies published in The Lancet demonstrate that Alzheimer's disease can be identified long before clinical symptoms appear. In a blood‑test investigation of 1,350 middle‑aged adults in the United States (average age 61), researchers measured two amyloid biomarkers and p‑tau217. Eighty‑six participants showed elevated levels, which were associated with slower processing speed, poorer executive function and a 2.5‑ to 4‑fold higher risk of rapid verbal‑memory decline over a five‑year follow‑up.

A separate PET‑imaging trial involving 682 participants in the United States and Canada compared the standard tau tracer Flortaucipir with the newer MK6240. MK6240 detected tau‑positive cases in more than twice as many individuals (15 % vs 6 % in cognitively normal, amyloid‑positive subjects) and identified additional cases among those with mild cognitive impairment.

Experts highlighted the potential of these tools to guide preventive strategies and enrol patients in disease‑modifying trials. Dr Jacqui Hanley (Alzheimer’s Research UK) called the findings “incredibly valuable,” while acknowledging that blood tests are not yet suitable for mass screening due to false‑positive risks. The studies suggest a path toward earlier diagnosis, which could affect millions of individuals at risk of dementia worldwide.
