Cancer immunotherapy advances with stem cell T-cells and mRNA vaccines

Recent advancements in cancer immunotherapy are offering new approaches to treating solid tumors. One research breakthrough involves a platform that generates T-cells from hematopoietic stem cells. Unlike current personalized therapies that require extracting and modifying a patient's own cells—a process that is often slow and costly—these lab-grown cells are designed to be ready-to-use. This method has shown promise in targeting tumors in organs such as the lungs and ovaries, demonstrating better persistence and a superior ability to reach tumor sites compared to traditional engineered T-cells.

In a separate development, a Phase III study has demonstrated the efficacy of a personalized mRNA vaccine named Intismeran, developed by Moderna. The vaccine is tailored to individual patients by sequencing the DNA of surgically removed tumors to identify specific mutations, or neoantigens. A bioinformatics algorithm then selects the most effective antigens to create a custom mRNA vaccine. This therapy aims to train the immune system to recognize and eliminate micrometastatic cells, specifically targeting high-risk melanoma to prevent recurrence and metastasis. The technology also shows potential for treating other difficult cancers, including kidney and bladder cancer.
