Melanoma mRNA vaccine reduces recurrence risk by 42%

A personalized mRNA cancer vaccine has demonstrated significant success in a phase 3 clinical trial, reducing the risk of melanoma recurrence or death by 42% compared to immunotherapy alone. The trial, known as INTerpath-001, involved 1,137 patients across 157 sites globally who had undergone surgery for high-risk melanoma.

Developed by Merck and Moderna, the vaccine, named intismeran, works by analyzing the unique genetic profile of a patient's tumor to teach the immune system to recognize and attack specific cancer cells. In the study, participants received pembrolizumab (Keytruda) alongside either the personalized vaccine or a placebo. After two years, 82% of those receiving the combination therapy were alive without the cancer returning, compared to 70% in the immunotherapy-only group.

Professor Georgina Long, a lead researcher from the Melanoma Institute Australia, described the results as a foundational shift in cancer treatment, marking a milestone in personalizing immunotherapy to prevent disease relapse.
