Daraxonrasib shows clinical promise for RAS-mutant lung cancer
Clinical trial data published in the New England Journal of Medicine indicates that daraxonrasib, an oral RAS inhibitor, shows promise in treating advanced non-small cell lung cancer (NSCLC) with RAS mutations. The drug, which was previously approved for pancreatic cancer, acts as a molecular glue by binding RAS with the protein cyclophilin A to block tumor growth signaling.
In a Phase 1/2 trial involving patients who had previously undergone platinum-based chemotherapy and immunotherapy, researchers observed significant results. In a specific cohort of 38 patients receiving doses between 160-220 mg, the objective response rate was 42%, with a median response duration of 11.5 months. The median progression-free survival was 8.3 months, and the median overall survival was 16 months.
These findings represent a potential improvement over the current standard of care, such as docetaxel regimens, which historically report response rates of only 9% to 14% and shorter progression-free survival. The therapy is currently moving forward in the Phase 3 RASolve 301 trial.
