Sacituzumab tirumotecan shows promise in treating advanced endometrial cancer

Results from the Phase 3 TroFuse-005 international clinical trial indicate that the experimental antibody-drug conjugate sacituzumab tirumotecan (sac-TMT) significantly improves outcomes for patients with advanced or recurrent endometrial cancer.

The study, presented at the European Society for Medical Oncology (ESMO) congress and published in The Lancet, involved 776 women across 242 centers in 31 countries. Patients in the trial had previously undergone platinum-based chemotherapy and anti-PD-1/L1 immunotherapy. Compared to standard chemotherapy (doxorubicin or paclitaxel), sac-TMT reduced the risk of death by 32% and the risk of disease progression or death by 41%.

Key findings include a median overall survival of 19 months for the sac-TMT group versus 12 months for the control group. Median progression-free survival was 7.2 months compared to 3.9 months. Additionally, the objective response rate nearly doubled, rising from 17.3% in the chemotherapy group to 35.1% in the sac-TMT group. The drug targets the Trop2 protein to deliver targeted chemotherapy directly to tumor cells. Common side effects noted included low blood counts and oral mucositis.
