Enteric Neurons Linked to Constipation and Abdominal Pain in New Mouse Study

Scientists have identified a specific group of neurons in the gut that appears to coordinate both the movement of fecal matter through the colon and the perception of abdominal pain. The neurons express the prodynorphin (Pdyn) gene and are concentrated within the enteric nervous system, often called the “second brain.”

When these cells were activated in isolated mouse colons, fecal pellets moved forward more readily. In live mice, the same activation produced behaviors associated with pain or discomfort. The findings suggest that a single population of gut neurons may drive both constipation and pain, providing a new target for future therapies. The study is posted as a bioRxiv preprint and has not yet undergone peer review.
