Genmab petosemtamab combination reduces head and neck cancer death risk by 50%

Genmab announced that its bispecific antibody petosemtamab, when used in combination with MSD’s Keytruda (pembrolizumab), met both primary endpoints in a Phase 3 trial for first-line treatment of PD-L1-positive recurrent or metastatic head and neck squamous cell carcinoma (HNSCC).

In the LiGeR-HN1 trial, the combination therapy reduced the risk of death by 50% compared to pembrolizumab alone, according to an interim analysis by an independent data monitoring committee. The study also showed a statistically significant improvement in the objective response rate, with an odds ratio of 3.17.

The clinical benefits were observed regardless of human papillomavirus (HPV) status. The combination was reported to be generally well tolerated, with no new safety signals identified. Petosemtamab is an investigational IgG1 bispecific antibody that targets EGFR and LGR5.

Following these results, Genmab shares saw significant increases. The company plans to present full results at an upcoming medical meeting and engage with global regulatory authorities regarding the therapy.
