GLP-1 receptor agonists linked to reduced fracture risk and chronic pain modulation

Recent research suggests that GLP-1 receptor agonists (GLP-1 RAs) may offer health benefits beyond weight loss and glycemic control. A study published in JAMA, utilizing data from the TriNetX Research Network, found that among adults aged 50 to 90 with type 2 diabetes, treatment with GLP-1 RAs was associated with a 21% reduction in the risk of fragility fractures over three years compared to those treated with DPP-4i. The most significant reductions were observed in vertebral fractures and hip or femur fractures.

Additionally, a review published in the journal Nutrients explored the potential of GLP-1 signaling to influence chronic pain management. Researchers suggest that GLP-1 RAs may affect nociception through the gut-brain axis and the gut microbiome. The review indicates that GLP-1 provides signals that regulate gastrointestinal, metabolic, and immune functions, potentially mitigating pain sensitization caused by gut dysbiosis and altered microbial signaling.
