GLP‑1 drugs linked to loss of pleasure, reduced binge‑eating and lower depression risk
Doctors have reported three cases of anhedonia – a loss of pleasure and motivation – in patients taking the highest doses of tirzepatide, a GLP‑1 medication used for obesity. All three women regained interest in daily activities after lowering the dose, and one required adjunct bupropion. The side effect appears limited to large doses and can be managed with dose adjustment.
A systematic review of 25 randomized trials found that GLP‑1 receptor agonists (including semaglutide, tirzepatide and liraglutide) significantly reduced binge‑eating episodes, loss‑of‑control eating and emotional eating, suggesting a therapeutic role for binge‑eating disorder. Researchers note the need for independent, long‑term trials and better public‑health access.
A drug‑target Mendelian randomization study using genetic proxies for GLP‑1 receptor activation reported lower odds of major depressive disorder, bipolar disorder and other mental‑health conditions, indicating possible broader psychiatric benefits of GLP‑1 activation. Clinical confirmation is still required.
