GPR133 receptor identified as potential osteoporosis treatment target
Researchers from Leipzig University and Shandong University have identified a potential biological target for treating osteoporosis. The study, published in Signal Transduction and Targeted Therapy, focuses on the GPR133 receptor, a member of the adhesion G protein-coupled receptor family.
In experimental models, mice with genetic impairments to GPR133 exhibited early-age bone density loss similar to human osteoporosis. To address this, scientists used a computer-assisted screen to identify AP503, a compound that acts as a stimulator for the GPR133 receptor.
Testing showed that AP503 increased bone strength in both healthy mice and those with osteoporosis-like bone loss. The mechanism appears to work by shifting the cellular balance: it encourages osteoblast activity (bone building) while reducing osteoclast activity (bone breakdown). While these results are promising, researchers noted that AP503 has not been tested in humans and significant further work is required before clinical applications can be considered.
