Karolinska Institutet studies link inflammation and tobacco to cluster headaches
Two new studies from Karolinska Institutet in Sweden have provided fresh insights into the biological mechanisms of cluster headache, a rare and severe neurological disorder. The research explores the roles of genetic factors, inflammation, and environmental exposures in the development of the condition.
One study, published in The Journal of Headache and Pain, analyzed genetic risk factors in over 1,500 individuals. Researchers confirmed the relevance of seven genes previously associated with the disorder, noting that six of them are active in immune cells or linked to inflammatory signaling pathways. This supports the hypothesis that the immune system and inflammation are central to the condition.
A second study, published in Cephalalgia, investigated the link between tobacco smoke and cluster headaches. Researchers found that patients exhibited higher activity in genes linked to nicotine exposure and heavy metal binding. The study noted that blood and spinal fluid from patients contained indirect markers associated with heavy metal exposure, such as cadmium, lead, and arsenic, alongside changes in DNA methylation. While the findings suggest that toxic metals in cigarette smoke may influence the biology behind the pain, researchers noted the results are indirect and based on a small sample size.
