Moderna and Merck mRNA melanoma vaccine shows success in phase 3 trial
A phase 3 clinical trial, known as INTerpath-001, has demonstrated that a personalized mRNA vaccine, intismeran, combined with the immunotherapy drug pembrolizumab, significantly improves outcomes for patients with high-risk melanoma. The study, conducted by Moderna and Merck, involved 1,137 patients and showed statistically significant improvements in both recurrence-free survival (RFS) and distant metastasis-free survival (DMFS).
The vaccine is highly personalized; it uses a computational algorithm to identify up to 34 specific neoantigens from a patient's own tumor tissue to instruct the immune system to target remaining cancer cells. Notably, the combination therapy did not show an appreciable increase in side effects or toxicity compared to using pembrolizumab alone.
The National Cancer Institute in Milan played a leading role in the research, enrolling 82 patients, which was the highest number of any single center in the international study. If further studies confirm the efficacy and safety of this approach, pharmaceutical companies aim to bring the personalized vaccine to market by 2027.
