Molecular mechanisms of retinal circuit development identified in mouse studies

Recent research into mouse retinal development has identified key molecular mechanisms that govern how neuronal types integrate into functional visual circuits.

One study highlights the role of Protocadherin 9 (Pcdh9), an autism-associated risk gene, in the survival of bipolar cells. Researchers found that the deletion of Pcdh9 leads to the loss of both OFF and ON cone bipolars, while disrupting its extracellular binding specifically affects ON cone bipolars through homophilic interactions with synaptic partners.

Complementary research focused on the establishment of the retinal OFF pathway. The study identified LRFN2, a cell-adhesion molecule expressed in cones, as a critical factor in stabilizing contacts with OFF bipolar-cell dendrites. LRFN2 organizes ionotropic glutamate receptor clusters to establish excitatory transmission. While the loss of LRFN2 abolishes direct dendritic excitation in OFF bipolar cells, these cells maintain contrast-dependent glutamate output via ON-driven axonal inhibition. This disruption was shown to weaken retinal responses to looming visual stimuli.
