Orelabrutinib and sintilimab combination shows efficacy in CNS lymphoma study

A prospective phase 2 study published in BMC Cancer indicates that combining the BTK inhibitor orelabrutinib with the anti-PD-1 antibody sintilimab produces high response rates in patients with relapsed or refractory primary central nervous system lymphoma (PCNSL).

The trial, conducted at four Chinese centers, enrolled 28 patients. The study reported an overall response rate of 60.7 percent after four cycles of treatment, which included complete responses in 28.6 percent of participants. At a median follow-up of 22.9 months, the median progression-free survival was 8.4 months, while the median overall survival had not yet been reached.

The treatment was described as well-tolerated with a manageable safety profile. Common adverse events included fatigue, bleeding, infection, leukopenia, and neutropenia. No unexpected toxicities were reported, and no immune-related adverse events occurred during the study.
