Osteoporosis linked to gum disease, missed fracture diagnoses, and novel drug target
A study by Griffith University (Australia) and Nanjing University (China) found that periodontitis heightens fracture risk in older adults with osteoporosis, aggravating bone loss in the jaw and femur and increasing systemic inflammation.
Research using Austrian health‑insurance data on more than 1.7 million adults aged 50‑79 showed that many fragility fractures—especially of the wrist, upper arm and pelvis—are not recognized as warning signs of osteoporosis, with hip fractures most strongly associated with a diagnosis. The analysis also highlighted a gender gap: women are more often diagnosed alongside other chronic conditions, while men, particularly those aged 50‑69, frequently receive a diagnosis only after a fracture and often have alcohol‑related health issues.
A critique of the current bone‑density screening system notes that the WHO‑endorsed T‑score thresholds, established in 1994 by a committee funded by pharmaceutical firms, classify about 80 % of women over 50 as osteopenic or osteoporotic, yet two‑thirds of fractures occur in individuals who do not meet these cut‑offs. Bisphosphonate drugs, originally developed for industrial water softening, raise density readings but can make bone more brittle, with fracture rates rising after three to five years of use.
Separate pre‑clinical work from Ulm University (Germany) identified the Axl receptor as a regulator of bone formation. Blocking Axl with the inhibitor BGB324 enhanced osteoblast activity and increased bone mass in mice, suggesting a potential new anabolic therapy for osteoporosis.
