Research identifies genetic and neuropathological drivers of dementia
Recent research has provided new insights into the neuropathological drivers of cognitive decline and the genetic architecture of dementia-related proteinopathies.
A study published in the Journal of Neuropathology & Experimental Neurology examined how concurrent neuropathologies affect cognitive decline in patients with Alzheimer disease. The findings indicated that vascular brain injury has a minimal impact on cognitive decline when compared to other combinations of neuropathologies.
In a separate study involving 966 participants, researchers utilized whole-genome sequencing and multidimensional modeling to identify genetic associations with specific proteinopathies, including Aβ, tau, α-synuclein, and TDP-43. The study validated known risk alleles, such as TMEM106B and GRN for TDP-43 pathology and GBA for α-synuclein, while also identifying novel suggestive alleles including SDHAF1, TMEM68, and ARHGEF28.
