Researchers Reveal Epigenetic Changes in Dopamine Neurons from Early Trauma

A study published in the journal Neuron shows that early‑life stress permanently alters the epigenetic landscape of dopamine‑producing neurons in the ventral tegmental area (VTA). The research, conducted by scientists at Washington University School of Medicine and Princeton University, found that childhood trauma raises levels of the enzyme SETD7, which adds the H3K4me1 tag to histones, causing DNA in these neurons to become less tightly coiled. This “uncoiling” lowers the activation threshold for stress‑responsive genes, making the neurons hyper‑reactive and leading to heightened anxiety‑ and depression‑like behaviors in adult mice.

When the researchers blocked SETD7 after early adversity, the chromatin remained compact and the mice were protected from stress‑induced social avoidance, suggesting a potential therapeutic target for mitigating long‑term mental‑health effects of early trauma.
