Weizmann Institute study finds Viagra may block cancer metastasis

Researchers at the Weizmann Institute of Science, led by Ayelet Erez and Yarden Ariav, published a study in Cancer Research showing that sildenafil (Viagra) can inhibit cancer metastasis. The drug blocks the enzyme phosphodiesterase‑5 (PDE5), raising intracellular cGMP levels. Elevated cGMP interferes with the NPC1 protein, reducing cholesterol transport inside tumor cells and limiting their ability to migrate and form new tumors.

Laboratory tests on human and mouse tumor cells demonstrated that this metabolic disruption makes highly migratory cancer cells especially vulnerable. The anti‑metastatic effect was further enhanced when sildenafil was combined with statins, which also lower cholesterol availability. A retrospective analysis of millions of patient records indicated higher survival rates for cancer patients who had used sildenafil, with an even stronger benefit for those also taking statins. The study involved Israeli and U.S. researchers and suggests a potential new therapeutic avenue, though clinical use has not yet been established.
