Synthetic foldamer SK-129 shows promise against Parkinson's disease

Researchers led by Sunil Kumar at the University of Denver have created a synthetic molecule called SK-129, a laboratory‑designed foldamer that binds to several regions of the protein alpha‑synuclein. By holding the protein in a shape that is less likely to stick to other copies, SK-129 prevents the formation and spread of toxic protein clumps that are implicated in Parkinson’s disease.

In mouse studies the compound reduced disease‑related symptoms, suggesting a potential new therapeutic avenue not only for Parkinson’s but also for other neurodegenerative disorders such as Alzheimer’s disease and ALS. Foldamers are noted for their ability to engage targets that traditional drugs cannot reach.

The molecule has not yet been tested in humans, and questions remain about its long‑term safety, optimal dosing and how it behaves in the human body. Additional research is required before clinical use can be considered.
