TPC1 Ion Channel Identified as Key Regulator of Iron Homeostasis
Researchers led by pharmacologist Professor Christian Grimm of Ludwig Maximilian University of Munich and the University of Oxford have shown that the ion channel TPC1 controls cellular iron balance. Using patch‑clamp recordings in mouse models, the team found that a genetic mutation that hyper‑activates TPC1 causes excessive iron accumulation, while complete loss of the channel leads to iron deficiency. The channel influences iron uptake by modulating endosomal pH and transferrin‑mediated transport. These findings provide new insight into the mechanisms behind common iron‑related disorders such as anemia and hereditary hemochromatosis and suggest that targeting TPC1 could become a therapeutic strategy for both iron overload and deficiency conditions.
