Travere Therapeutics reports sparsentan reduces biomarkers in IgA nephropathy
A biomarker analysis from the phase 3 PROTECT trial indicates that sparsentan (Filspari) reduces urinary biomarkers of macrophage, B-cell, and complement activation, as well as inflammation, in adults with IgA nephropathy (IgAN).
Sparsentan, a dual endothelin and angiotensin receptor antagonist, was compared against irbesartan over a 110-week period. The analysis showed reductions in urinary sCD163, BAFF, C5b-9, and IL-6. In the PROTECT trial, sparsentan reduced proteinuria by 49.8% compared to 15.1% with irbesartan at week 36 and improved the chronic 2-year eGFR slope by approximately 1.1 mL/min/1.73 m² per year.
Travere Therapeutics presented these findings at ASN Kidney Week 2026. The results build upon exploratory signals previously observed in the smaller SPARTAN study.
