tRNA modifications drive Chagas parasite transformation

Researchers have identified that chemical changes in transfer RNA (tRNA) drive the transformation of the Chagas disease parasite, Trypanosoma cruzi, during its life cycle. This process allows the parasite to adapt its protein production to transition from a non-infectious state to an infectious phase capable of infecting humans and other mammals.

The study, published in PLOS Pathogens, involved mapping 170 modification sites in tRNA molecules using tRNA sequencing, mass spectrometry, and bioinformatics. Lead researcher Herbert Guimarães de Sousa Silva, who conducted the work at the Federal University of São Paulo and the Butantan Institute, noted that these modifications act as regulators for the delivery of amino acids essential for protein assembly.

To further investigate the impact of these chemical alterations, the team utilized CRISPR gene-editing technology to evaluate how the absence of specific modifications affects the parasite's ability to transition between life cycle phases.
