UCLA study finds rapamycin briefly reverses autism-like traits in mice

Researchers at UCLA Health reported that a single dose of the immunosuppressant rapamycin rapidly improved autism-like brain dysfunction and behavioral traits in adult mice. The study, published in Nature Communications, showed reductions in brain overactivity, seizure risk, sensory sensitivity, and repetitive behaviors within hours of treatment. However, the benefits were short‑lived, with symptoms returning after about 72 hours and repeated dosing quickly losing efficacy due to drug tolerance and toxicity. Senior authors Dr. Harley Kornblum and Dr. Janel Le Belle emphasized that the findings suggest adult brain circuitry remains adaptable, shifting focus from structural changes to functional pathways such as the mTOR signaling cascade. They cautioned that rapamycin itself is unsuitable for human autism therapy because of safety concerns, and called for development of safer methods to modulate neuronal circuits.

The research used an outbred CD1 mouse model exposed to mild inflammatory stimuli during pregnancy, which produced persistent autism-like phenotypes. While the results offer a proof of concept for targeting functional brain networks, translation to clinical treatment will require alternative compounds with lower toxicity.
