University of California researchers identify molecule to prevent muscle loss during weight loss

Current anti-obesity pharmacological treatments, such as those based on semaglutide, are effective for weight loss but often result in the loss of muscle mass alongside fat. Research from the University of California, Berkeley, published in Science Advances, has identified a potential solution using a molecule called TOFA (5-tetradecyloxy-2-furoic).

Unlike traditional treatments that focus on caloric intake, TOFA works by increasing caloric expenditure. The molecule blocks cellular fat production while activating receptors (PPARα and PPARδ) that encourage cells to burn fat for energy. In studies involving obese mice, TOFA improved insulin sensitivity, lowered triglycerides, and reduced signs of fatty liver without impacting muscle tissue.

When administered alongside semaglutide or tirzepatide, TOFA demonstrated a synergistic effect, producing superior weight loss and glycemic control compared to using either drug alone. This approach aims to increase cellular energy consumption by up to 18% without additional physical movement.
