Washington University researchers identify brain mechanism to control chronic pain

Researchers at the Washington University School of Medicine in St. Louis have identified a biological mechanism in the brain that could potentially control chronic neuropathic pain. The study, published in ‘Current Biology’, focuses on the locus coeruleus, a brain region responsible for alertness, stress, and pain processing.

In healthy conditions, specific mu-opioid receptors in this region act as a natural ‘brake’ to suppress pain signals traveling up the spinal cord. However, the research found that nerve damage can cause this system to become hyperactive, contributing to persistent pain rather than suppressing it. Through experiments on mice, scientists observed that removing these receptors increased sensitivity to touch and temperature, while restoring them eliminated the hypersensitivity.

This discovery offers a potential pathway for developing more precise therapies. Unlike traditional opioid medications, which affect receptors throughout the entire body and carry risks of addiction, tolerance, and side effects, future treatments could specifically target the receptors within the locus coeruleus to manage pain more safely.
