Yale researchers identify genetic mechanism for kidney transplant rejection
Researchers at the Yale School of Medicine have identified a potential genetic mechanism that increases the risk of kidney transplant rejection, particularly in individuals of recent African ancestry. The study, published in the Journal of Clinical Investigation, focuses on two variants of the apolipoprotein L1 (APOL1) gene, known as G1 and G2.
Led by Madhav Menon, the research team used genetically engineered mouse models to isolate the biological effects of these variants. The findings revealed that mice carrying the G1 or G2 variants exhibited heightened activation of T cells, which are immune cells central to the organ rejection process.
While APOL1 variants have previously been linked to kidney disease, this research provides new insight into how they specifically contribute to transplant rejection through altered immune cell signaling. This discovery may help address health disparities, as African Americans represent approximately 36% of people on dialysis in the United States despite making up 13% of the population.
