Recent clinical developments have highlighted the efficacy of Bruton tyrosine kinase (BTK) inhibitor combinations in treating various forms of lymphoma. A phase 2 study conducted at four Chinese centers reported that combining orelabrutinib with the anti-PD-1 antibody sintilimab produced high response rates in patients with relapsed or refractory primary central nervous system lymphoma. The study, published in BMC Cancer, noted an overall response rate of 60.7 percent and described the treatment as well-tolerated with a manageable safety profile. In the United States, the FDA has granted priority review to a supplemental new drug application from BeOne Medicines for zanubrutinib BRUKINSA (zanubrutinib) in combination with rituximab for adults with previously untreated mantle cell lymphoma. lymphoma (MCL). This decision follows the phase 3 MANGROVE trial, which involved 510 patients across 157 global sites. The trial indicated that the combination significantly improved progression-free survival compared to bendamustine plus rituximab, reducing the risk of disease progression or death by 43 percent. If approved, this would offer a chemotherapy-free, BTK inhibitor-based regimen for frontline MCL treatment. Full results from the MANGROVE study are expected to be presented at the 68th American Society of Hematology Annual Meeting in December 2026.
