Advancements in MYC cancer protein research
Overview
Recent medical research has focused on addressing the MYC protein, a master regulator of cell growth whose deregulation is linked to a majority of human cancers. Because the protein lacks traditional binding pockets, it has historically been classified as ‘undruggable’.
Researchers at the Sanford Burnham Prebys Medical Discovery Institute identified a regulatory hub at the PVT1 locus on human chromosome 8q24. Their studies indicate that PVT1 encodes two novel proteins essential for MYC activity in cancer cells.
Following this, scientists at MD Anderson Cancer Center reported the development of a preclinical therapy, GT19630. This drug targets a ‘feedforward loop’ between the MYC protein and the GSPT1 protein. By binding to both, the therapy marks MYC for disposal and degrades GSPT1. The treatment has shown anti-cancer activity in models of leukemia, lymphoma, and multiple myeloma, including treatment-resistant cases.
