Alzheimer’s disease genetic and genomic research
Overview
Scientific research into Alzheimer’s disease has expanded from identifying specific genetic risk factors to understanding complex genomic structures. Initial findings highlighted how rare loss-of-function variants in the PLCG2 gene can increase disease risk tenfold. Additionally, studies on the APOE4 gene demonstrated that the protein Nell2 causes neuronal shrinkage and abnormal memory circuit activity, though suppressing this protein showed potential for reversing early changes in mouse models.
Subsequent research has identified a new dimension of the disease involving the three-dimensional organization of the genome. Studies indicate that the physical folding of DNA in certain brain cells is altered in patients, which affects gene accessibility and tissue organization.
Recent investigations have further expanded this scope, suggesting that the three-dimensional organization of the genome varies in specific brain cells, offering a new regulatory layer for potential treatments. Researchers are also exploring the role of the immune system and peripheral tissues, noting that genetic signals associated with the disease are present in the lungs, intestines, and skin. This indicates that peripheral inflammation and immune processes may influence brain health. Furthermore, studies into neuroimmune dysregulation are examining how modulating immune proteins, such as PD-1, might restore microglial functions and improve communication between microglia and neurons.
Alongside these biological discoveries, medical experts have issued cautions regarding the clinical utility of commercial genetic testing. Experts note that strictly hereditary Alzheimer’s accounts for less than 1% of cases, while the remaining 99% stem from a complex interplay of environmental and genetic factors, making predictive genetic panels less effective for the general population.
