Overview
BioNTech reported that its experimental mRNA cancer immunotherapy, BNT113, failed to meet the primary endpoint in a combined Phase 2/3 clinical trial known as AHEAD-MERIT. The study evaluated the combination of BNT113 and Pembrolizumab (Keytruda) for patients with advanced, inoperable HPV16-positive head and neck squamous cell carcinoma.
The results indicated that the combination therapy did not demonstrate a statistically significant improvement in progression-free survival (PFS) compared to treatment with Pembrolizumab alone. Following the announcement, BioNTech’s US-listed shares declined by nearly 3% in early trading.
Despite the setback, the U.S. Food and Drug Administration (FDA) has granted BNT113 Fast Track designation. An independent data monitoring committee has recommended that the trial continue to evaluate the second primary endpoint: overall survival (OS). Researchers noted that because immune-mediated antitumor effects can build and deepen over time, achieving a statistically significant OS benefit remains critical to the study’s success and potential regulatory approval.
