Cancer treatment resistance research developments

Overview

Medical research has focused on identifying cellular mechanisms that contribute to cancer treatment resistance, particularly in prostate and liver cancers.

Initial studies at the University of Texas MD Anderson Cancer Center found that prostate tumors may use cancer-associated fibroblasts to create physical barriers that prevent T cells from infiltrating tumors. This finding may explain the limited efficacy of certain immunotherapies. Additionally, researchers at Texas A&M University identified enzymes that help liver cancer cells survive chemotherapy by activating a stress-protection system. Laboratory experiments suggested that combining specific enzyme blockers with cisplatin could increase the effectiveness of chemotherapy in killing liver cancer cells.

Subsequent research has further explored these resistance mechanisms. A meta-analysis of Phase-3 studies indicated that while adding hormone therapy to postoperative radiotherapy does not improve overall survival for all patients, those with higher PSA levels prior to radiotherapy may see specific benefits. Furthermore, researchers at the Fred Hutch Cancer Center identified that transfer RNA, specifically tRNA1Arg(UCU), acts as a molecular regulator in androgen receptor-dependent tumors. The study noted that while this molecule is concentrated in sensitive cells, it is significantly reduced in therapy-resistant tumors, suggesting that restoring tRNA levels could potentially re-sensitize resistant cells to androgen receptor inhibitors.
