Colorectal cancer research on age-linked biological traits
Overview
Researchers from the Dana-Farber Cancer Institute, Mass General Brigham, and the Broad Institute have identified distinct biological differences in colorectal tumors based on patient age. A study published in Nature Medicine utilized single-nucleus RNA sequencing to analyze 189 primary tumors, creating an atlas of over 850,000 nuclei.
The findings indicate that among microsatellite-stable tumors, those from younger patients contain proportionally more supporting tissue and fewer epithelial cells than those from older patients. Researchers also observed age-associated changes in gene activity, specifically an increased expression of long non-coding RNAs in the tumor epithelial cells of younger individuals and increased activity in genes associated with cancer cell mobility.
These discoveries follow data from the American Cancer Society showing that colorectal cancer diagnoses in individuals under 50 in the United States increased by approximately 2.9 percent annually between 2013 and 2022. The research, which was later confirmed through the analysis of an additional 1,276 tumors, provides a roadmap for investigating why the disease may behave more aggressively in younger populations.
