Research led by Columbia University and published in Nature Medicine indicates that major depressive disorder (MDD) is linked to stalled neurogenesis in the hippocampus. By analyzing approximately 500,000 brain cells, researchers found that while the brain continues to produce neural stem cells, the development of these cells into functional, mature neurons is functionally compromised. Studies show an accumulation of early-stage, quiescent, stem-like cells and a significant reduction in mature neurons. Molecular analysis identified dysregulation in genes related to synaptic transmission, cellular metabolism, and energy transport, alongside heightened markers of neuroinflammation and immune-related gene activity. These findings suggest a shift in the scientific understanding of depression, moving away from the traditional monoamine hypothesis focused on neurotransmitter imbalances toward a model of impaired cellular adaptability and structural plasticity. This disruption in cellular maturation may provide explanations for clinical symptoms such as cognitive rigidity, impaired memory, and difficulties with pattern separation. Scientists suggest these molecular signatures could eventually facilitate more precise, personalized treatments aimed at restoring the brain's ability to form new neurons.
