Development of glucose-responsive probiotics for diabetes
Overview
Researchers from East China Normal University have developed an engineered probiotic, named GIFT, designed to treat diabetes through a “sense and respond” mechanism. Published in the journal Nature, the study describes how the probiotic uses a synthetic gene circuit and the HexR regulatory protein to act as a molecular switch. When glucose levels rise, the system triggers the production of GLP-1 analogues, such as semaglutide, to lower blood sugar; production ceases once levels normalize.
In animal trials, the system showed efficacy in mice by improving insulin sensitivity and reducing organ damage. In cynomolgus monkeys with type 2 diabetes, oral administration maintained stable blood glucose levels over five weeks without toxic or inflammatory reactions. While these results are promising for mitigating side effects of long-term semaglutide use, further research is necessary to confirm efficacy in humans.
