DNA Typewriter technology for and PEtracer map mouse embryo lineage mapping lineages
Researchers from the University of Washington, Dartmouth College, and Sloan Kettering have developed a novel genetic marking technology called ‘DNA Typewriter’ to map the cellular lineage of mouse embryos. The technology works by inserting a synthetic DNA strip into a fertilized egg’s genome. As cells divide, a gene-editing technique adds sequential molecular barcodes to the strip, creating a permanent logbook or ‘flight recorder’ of developmental events. This allows scientists to reconstruct the family tree of approximately 1.3 million cells, tracking how they transform into specialized tissues and organs during gestation. This method addresses the difficulty of observing continuous mammalian development within the uterus. By preserving a running log of division events without fully severing the DNA double helix, the technology provides a high-resolution view of cell fate decisions during organogenesis. Recent studies published in Cell and Science have demonstrated the successful reconstruction of a high-resolution cellular family tree for a mouse embryo, tracking approximately 1.28 million cells over 13.5 days of gestation. The DNA Typewriter method utilizes CRISPR-based prime editing to ‘stamp’ genetic barcodes onto the DNA. In a parallel advancement, researchers from the Whitehead Institute at MIT and the Max Planck Institute for Molecular Genetics utilized a technique called PEtracer. This method introduced heritable genetic marks to resolve roughly 75 percent of cell divisions across more than 1.4 million cells from 16 mouse embryos between days 7.5 and 10 of gestation. Together, these tools may help researchers understand the origins of cancers, neurodevelopmental disorders, and congenital defects.
