Research into metabolic weight management is expanding through new pharmacological approaches that differ from existing GLP-1 agonists. Biotechnology company Enveda has reported that its experimental oral drug, ENV-308, successfully cleared a Phase 1 clinical trial involving 88 healthy adults. adults with overweight or obesity. The drug is designed using AI to mimic the biochemical effects of physical activity by targeting Lac-Phe, a molecule that signals the brain to reduce appetite. While the natural molecule disappears quickly, ENV-308 is formulated as a once-daily pill intended to remain in the body longer. Although the trial focused on safety rather than efficacy, researchers noted an “exceptional gastrointestinal safety profile” and suggested the drug may cause fewer adverse effects, such as nausea or vomiting, compared to existing GLP-1 therapies like semaglutide. Researchers intend to move toward Phase 2 trials to test the potential for weight maintenance. In parallel, scientists at the University of California, Berkeley, are investigating the compound 5-tetradecyloxy-2-furoic acid (TOFA). Animal studies suggest TOFA may improve glucose levels and insulin sensitivity by affecting fat production and cellular energy mechanisms without significant loss of lean mass.
