IDH1 Glioma Vaccine Shows 8-Year Survival; New Core2Edge Model Maps Tumor Infiltration
A phase‑1 clinical trial of an experimental vaccine targeting the IDH1 mutation in gliomas enrolled 33 patients with newly diagnosed high‑grade astrocytomas. When combined with standard treatment, the vaccine led to two‑thirds of participants still alive eight years after enrollment and a 42 % rate of disease‑free survival. Stronger immune responses, both T‑cell and B‑cell activity, correlated with the best outcomes, and booster doses given years later further heightened immunity without added side effects, suggesting a pathway toward long‑term tumor control.
Researchers at University Hospital Bonn and the University of Bonn introduced Core2Edge, a human‑tissue model that merges patient‑derived glioblastoma organoids with brain‑slice cultures discarded during neurosurgery. The platform visualises tumour cell invasion from the core into surrounding brain tissue using light‑sheet fluorescence microscopy, enabling drug testing directly on human tissue and eliminating the need for animal experiments. The method was published in Nature Protocols and offers a new avenue for studying aggressive brain‑tumour infiltrative behavior.
