Rentosertib AI-designed drug shows potential to reverse biological aging

A study published in Nature Biotechnology reveals that rentosertib, an AI-designed drug candidate from Insilico Medicine, may have the potential to reverse biological aging markers. Originally developed to treat idiopathic pulmonary fibrosis (IPF), the drug was evaluated in a Phase IIa clinical trial involving 42 patients.

Researchers utilized six independent proteomic aging clocks to analyze longitudinal serum data. All six models consistently showed a reduction in predicted biological age among patients treated with rentosertib compared to the placebo group. In some models, the reduction in biological age was estimated to be between 3 and 6 years.

In addition to aging markers, the study observed dose-dependent improvements in Forced Vital Capacity (FVC), a key indicator of lung function. The drug, which targets the TNIK kinase, was identified using the PandaOmics platform and designed via the Chemistry42 generative AI system. Rentosertib has now progressed to Phase III clinical trials.
