Overview
Insilico Medicine has developed an AI-designed drug candidate, rentosertib, which has shown potential in reducing biological aging markers during clinical trials. Originally intended to treat idiopathic pulmonary fibrosis (IPF), the drug targets the TNIK kinase.
In a Phase IIa clinical trial involving 42 patients, researchers used six independent proteomic aging clocks to analyze serum data. The models consistently indicated a reduction in predicted biological age among treated patients compared to a placebo group, with some models estimating a reduction of between 3 and 6 years. The most significant signals were observed around the fourth week of treatment. The study also noted dose-dependent improvements in Forced Vital Capacity (FVC), a measure of lung function. Specifically, patients in the 60mg once-daily dosage group saw a mean improvement of 98.4 milliliters, while the placebo group experienced a 20.3 milliliter decline.
Insilico Medicine utilized artificial intelligence for a dual purpose: identifying the TNIK enzyme as a target linked to both fibrosis and aging, and optimizing the drug molecule itself. While the results are promising for therapies addressing both specific diseases and the broader aging process, researchers cautioned that the study was small and that these measurements do not yet prove the drug can extend human lifespan or permanently reverse the aging process. Rentosertib has progressed to Phase III clinical trials.
