Rentosertib, an AI-designed drug, shows potential to reduce biological aging

An experimental drug, rentosertib, designed by Insilico Medicine using artificial intelligence, has demonstrated the ability to influence biological aging markers in patients with idiopathic pulmonary fibrosis (IPF).

In a Phase IIa clinical trial involving 42 participants, researchers utilized six different proteomic aging clocks to analyze blood samples. All six models detected a reduction in predicted biological age among the treatment group, whereas the placebo group showed little to no change. The most significant signals were observed around the fourth week of treatment. Depending on the specific model used, the shift in biological age was estimated to be between three and six years.

Insilico Medicine employed AI for a dual purpose: identifying the TNIK enzyme as a target linked to both fibrosis and aging, and optimizing the drug molecule itself. Beyond aging markers, the drug showed efficacy in treating lung disease; patients in the 60mg once-daily dosage group saw a mean improvement in forced vital capacity of 98.4 milliliters, compared to a 20.3 milliliter decline in the placebo group.

While the results are promising for developing therapies that address both specific diseases and the broader aging process, researchers note that the study was small and these findings do not yet prove that the drug can permanently reverse human aging or extend lifespan.
