KRAS mutation research reveals mechanisms of lung cancer drug resistance

Research into lung cancer resistance has identified two primary mechanisms by which tumors evade drugs targeting KRAS mutations. A study using mouse models revealed that some tumors respond to treatment by increasing the number of KRAS gene copies, thereby intensifying growth signals to overcome pharmacological blocks.

Alternatively, some tumors undergo a cellular identity shift, transitioning from adenocarcinomas to squamous cell carcinomas. This change reduces their dependency on KRAS, allowing them to grow via alternative, unidentified signaling pathways.

These findings complement the broader field of precision medicine, which seeks to move away from uniform treatment protocols toward personalized strategies. By analyzing the specific molecular and genetic characteristics of a tumor, specialists aim to identify ‘actionable’ alterations that can be targeted with specific therapies. However, the biological diversity of cancer means that even tumors appearing similar under a microscope can respond differently to treatment, and tumors can evolve over time, necessitating continuous molecular monitoring.
