Medical research reports divergent clinical trial results
Recent clinical trial results have yielded contrasting outcomes in medical research. In oncology, Phase II results from the I-SPY2 adaptive platform indicate that a combination of two immunotherapies, cemiplimab and fianlimab, administered before surgery alongside standard chemotherapy, significantly increased pathological complete response (pCR) rates in high-risk HER2-negative breast cancer patients. In the HER2-negative group, pCR rates rose from 21% to 44%, while triple-negative breast cancer rates increased from 29% to 53%. However, researchers noted excessive immunological toxicity, meaning this specific regimen will not proceed in its current form.
In cardiovascular research, the ZEUS clinical trial involving over 6,300 participants failed to show that the anti-inflammatory drug ziltivekimab reduces the risk of heart attack or stroke. Despite successfully reducing inflammation (measured by C-reactive protein) by blocking interleukin IL-6, the drug did not alter cardiovascular event rates or mortality. This outcome may challenge the long-standing theory that chronic inflammation is a primary independent risk factor for cardiovascular events, or suggest that the specific inflammatory pathway targeted was insufficient to influence clinical outcomes.
