Pancreatic Cancer Breakthroughs: KRAS inhibitor daraxonrasib and Vitamin D analog improve outcomes
Researchers reported two promising advances against pancreatic cancer, a disease that affects over 90% of patients with KRAS mutations and has a five‑year survival rate of about 12%. In a recent clinical study, the experimental KRAS‑targeting drug daraxonrasib more than doubled median survival for patients with advanced disease, extending it to 13.2 months versus 6.7 months on standard chemotherapy.
A separate trial examined the synthetic vitamin D analog paricalcitol, which reprograms fibroblasts that form a dense protective barrier around pancreatic tumors. When combined with standard chemotherapy, the drug increased partial response rates to 42% (versus 9% with chemotherapy alone), boosted immune‑cell infiltration, and prolonged progression‑free survival. Both approaches are still under investigation but suggest new strategies that could significantly extend survival and improve treatment effectiveness for pancreatic‑cancer patients.
